The kits utilize synthetic peptides which mimic the optimal substrate for distinct caspases and are labeled with either 7-amino-4-trifluoromethyl coumarin (AFC, fluorometric assay) or p-nitroanilide (pNA, colorimetric assay). Activated caspases in apoptotic cells cleave specifically their synthetic substrates resulting in release of free AFC or pNA, which is then quantified using a fluorometer or spectrophotometer, respectively. The kits are fast, simple, straightforward, and can be adapted for 96-well plates.
